About Retatrutide

Retatrutide (LY3437943) is an investigational triple agonist of the GLP-1, GIP and glucagon receptors, supplied as a lyophilized research peptide in 10mg and 30mg vials. It is studied in metabolic research for its effects on appetite signalling, glucose regulation and energy expenditure.

What is retatrutide?

Retatrutide, also known by its development code LY3437943, is a synthetic peptide developed by Eli Lilly and Company. It is a single molecule designed to activate three hormone receptors involved in metabolic regulation: the glucagon-like peptide-1 (GLP-1) receptor, the glucose-dependent insulinotropic polypeptide (GIP) receptor and the glucagon receptor.

Because it combines all three activities in one compound, retatrutide is often called a triple agonist or triple hormone receptor agonist. It has become one of the most closely followed compounds in incretin research, and it remains an investigational compound that is not approved by Health Canada or the FDA.

How retatrutide works

GLP-1 and GIP are incretin hormones released from the gut after nutrient intake. Their receptors, found in the pancreas, brain and other tissues, are involved in insulin secretion, appetite signalling and gastric emptying. Glucagon receptor activation is associated with hepatic glucose output, lipid metabolism and energy expenditure.

Retatrutide lets researchers study how incretin signalling and glucagon signalling interact when activated together. In its discovery study, the compound showed balanced activity at the GLP-1 and glucagon receptors with comparatively stronger activity at the GIP receptor, and its long half-life supported once-weekly administration intervals in clinical studies.

What published research has examined

Retatrutide was first characterised by Coskun and colleagues, who described its pharmacology at all three receptors along with its effects on body weight and glycaemic markers in preclinical models and early clinical testing (Coskun et al., Cell Metabolism, 2022).

A 48-week phase 2 trial in adults with obesity reported dose-dependent reductions in body weight (Jastreboff et al., New England Journal of Medicine, 2023). A parallel phase 2 trial examined glycaemic control and body weight in people with type 2 diabetes (Rosenstock et al., The Lancet, 2023).

Phase 3 studies in the TRIUMPH program have since reported results in obesity, type 2 diabetes and related conditions. That clinical literature is cited here for scientific context only and makes no claim about this research product.

Key research areas

  • GLP-1, GIP and glucagon receptor pharmacology
  • Appetite signalling and food-intake regulation
  • Glucose regulation and insulin secretion
  • Energy expenditure and lipid metabolism
  • Liver fat and cardiometabolic markers
  • Comparisons with single and dual incretin agonists

Retatrutide FAQ

How is retatrutide different from tirzepatide and semaglutide?

All three are incretin-based peptides, but they act on different receptor combinations. Semaglutide is a GLP-1 receptor agonist, tirzepatide is a dual GIP and GLP-1 receptor agonist, and retatrutide adds glucagon receptor activity to become a triple GLP-1, GIP and glucagon receptor agonist.

What retatrutide sizes does BioAscend carry?

Retatrutide is available as lyophilized powder in 10mg and 30mg vials, shipped from Canada.

How should retatrutide be stored?

Store unopened vials in a cool, dry place away from direct light, with refrigeration recommended for long-term storage. After reconstitution, keep the vial refrigerated at 2–8°C.

Is retatrutide approved for human use?

No. Retatrutide is an investigational compound that has not been approved by Health Canada or the FDA. BioAscend supplies it strictly for laboratory research use.
