5-Amino-1MQ (5-amino-1-methylquinolinium) is a small-molecule inhibitor of the enzyme nicotinamide N-methyltransferase (NNMT), supplied in 50mg vials. It is studied in research on adipose tissue metabolism, NAD+ pathways and cellular energy regulation.
5-Amino-1MQ is a quinolinium compound: a small molecule rather than a peptide. It belongs to a series of methylquinolinium analogues designed as selective, membrane-permeable inhibitors of nicotinamide N-methyltransferase (NNMT), with a reported half-maximal inhibitory concentration in the low micromolar range.
Its small size and cell permeability make it a useful tool compound for studying what happens inside cells when NNMT activity is reduced.
NNMT transfers a methyl group from S-adenosylmethionine (SAM) to nicotinamide, a form of vitamin B3. The enzyme is highly expressed in the liver and in white adipose tissue, and higher NNMT expression has been associated with obesity and insulin resistance in research models.
Because NNMT consumes both nicotinamide and SAM, its activity can affect the cellular NAD+ salvage pathway and the methylation balance of the cell. Inhibiting NNMT with 5-Amino-1MQ lets researchers study how these linked pathways influence lipid storage and energy expenditure in fat cells.
In the study that introduced this compound series, selective NNMT inhibitors reduced lipogenesis in cultured adipocytes. In mice with diet-induced obesity, a lead inhibitor reduced body weight, white adipose mass and adipocyte size and lowered plasma cholesterol without changing food intake (Neelakantan et al., Biochemical Pharmacology, 2018).
